ATP concentration should be greater in cells treated with DNP because more oxidation of glucose is correlated with increased ATP synthesis.

Dinitrophenol (DNP) is a chemical that acts as an uncoupling agent, making the inner mitochondrial membrane leaky to protons.

More glucose gets broken down in DNP-treated cells. This means that the rate of glucose oxidation increases in the presence of DNP. Dinitrophenol disrupts the H+ gradient reducing ATP synthesis. Under these conditions, much of our food that we eat could not be used for ATP synthesis are we lose weight.

DNP allows hydrogen protons to cross the membrane freely. Therefore there is no true proton gradient and while ATP production can still occur (some hydrogens can still go through ATP synthase), it will be at a much lower rate and much less efficient.
