The hydrolysis of pyrophosphate to orthophosphate is important in driving forward biosynthetic reactions, such as the synthesis

of DNA. This hydrolytic reaction is catalyzed in E. coli by a pyrophosphate that has a mass of 120 kd and consists of six identical subunits. For this enzyme, a unit of activity is defined as the amount of enzyme that hydrolyzes 10 umol of pyrophosphate in 15 min at 37 degrees C under standard assay conditions. The purified enzyme has a Vmax of 2800 units per milligram of enzyme.
a) How many moles of the substrate is hydrolyzed per second per milligram of enzyme when the substrate concentration is much greater than Km? (Draw a velocity versus substrate
concentration graph to show your point)

b) How many moles of active sites is there in 1 mg of enzyme? Assume that each subunit has
one active site.

c) What is the turnover number of the enzyme? (Don't bother comparing it to other enzymes;
just show all your own, which equations you use, what numbers you use, where you get them
from, etc.)

Answers:

a) How many moles of the substrate is hydrolyzed per second per milligram of enzyme when the substrate concentration is much greater than Km?

Answer:

31.1 µmol PPi s-1 mg -1

b) How many moles of active site are there in 1 mg of enzyme?

Answer:

5.0 x 10-8 mol

c) What is the turnover number of the enzyme?

Answer:

3732 s-1 per mol enzyme but there are 6 active sites per enzyme therefore turnover is 622 s-1 (per active site)

Explanation:

i have attached two pictures that explains the methodology as i can not write formulas here that's why i solved it and created a picture for attachment.

Are the counts of blue crabs in Maryland accurate for 1991 and 1999?
kirill [66]
The population of blue crabs will be difficult for the scientists to count because of unreliable ways and natural phenomenon or occurrence that causes the water's temperature to rise. According to climate reports, year 1999 have warmer temperature and this probably caused the blue crabs to be active, but on year 1990 female blue crabs sustained a normal quantity of production of their species. Thus, the estimated count of population of blue crabs is probably more accurate in year 1990 than year 1999 because of the stable production of the female crabs.

I need 10 interesting facts about Oviraptor? Please
Norma-Jean [14]
Oviraptor means "egg theif"

Oviraptors could have 20 eggs at a time

They lived in Mongolia

The aren't necessarily true raptors

Oviraptors were about 6 - 8 feet tall

The first oviraptor was found in 1924 (i believe)

Oviraptors lived in the Cretaceous period.

Oviraptors most likely weighed 60 - 70 pounds.

They were renamed, as "ovinutrix"

The first fossil of oviraptors was found with eggs.

What is mitosis and why is it important for identical cells to be produced
Ksivusya [100]

Mitosis is a type of cell division that results in two daughter cells each having the same number and kind of chromosomes as the parent nucleus, typical of ordinary tissue growth. Mitosis is important because the organism must:

1. replace cells that have worn out or died

2. repair tissue that is damaged

3. add cells in order to grow

4. make new kinds of cells in order to develop
