A student isolates a protein from anaerobic bacteria and analyses the protein by polyacrylamide gel electrophoresis containing S

DS (PAGE-SDS). Following protein staining, a single band appears, which excites the student's supervisor. To be certain, the supervisor suggests that the student run a second electrophoresis under native conditions (i.e. non-denaturing, or without SDS). This gel shows two bands after staining. Assuming no errors were committed during these experiments, explain the observations.
A. In the absence of SDS, protein subunits separate into two peptide with charges, leading to two peaks.
B. In the absence of SDS protein subunits separate into two peptides with different masses, leading to two peaks.
C. In the presence of SDS two proteins with identical charges but different molecular weights will show a single band, while in the absence of SDS different molecular weights will create separation.
D. In the presence of SDS two proteins with identical molecular weight but different isoelectric points will show a single band, while in the absence of SDS different charges will create separation.

Answer:

The correct answer is option D.

Explanation:

In the presence of SDS single band appeared, while in the absence of SDS two bands appeared. SDS or sodium dodecyl sulfate refers to an anionic detergent, which combines with amino acid side chains providing the protein a net negative charge. It dissociates the non-covalent bonds.

In SDS-PAGE, the separation of proteins takes place on the basis of their molecular weight. Options A and B are incorrect as only in the presence of SDS, the separation of protein subunits takes place. Option C is also incorrect as a protein containing distinct molecular weight cannot show single band.

Option D is correct as the presence of SDS supplements a bunch of negative charges to the protein, thus, charge is not the factor. Therefore, the proteins are distinguished on the basis of the molecular weight. Thus, identical molecular weight demonstrates a single band. In the non-presence of SDS, charge performs a function along with the molecular weight, therefore, two bands appear.

How do scientists obtain knowledge about the world?
Sphinxa [80]
Well, scientists depend on science.They use the process of observation. But there are processes for these. The processes are;
Asking a question; Scientists wonder how specific happenings can be explained, they look for an answer.
They perform research; the scientists study the particular thing, and look for reasons for a particular happening.
They form a hypothesis; a hypothesis is a statement which is not universally true, they are observations that haven't been agreed upon.
They test their hypothesis; they perform experiments to find out if their hypothesis is true.
They analyse the data and draw a conclusion; they record their hypothesis and write down their end result.
They pass across their result; they pass their new observation across to other people.

The effect of cholesterol on membrane fluidity at physiological temperatures is to Choose one: A. prevent lateral movement of ph
kobusy [5.1K]

Answer:

The correct answer is D. maintain membrane fluidity through its disruption of fatty acid packing.

Explanation:

Cholesterol is a steroid lipid and is a constituent of biological membranes. It regulates the fluidity of the membrane (so, option C is not correct). Since cell membranes are composed of another type of lipids, the phospholipids, which form a bilayer, cholesterol distributes between the phospholipid tails and avoids these molecules pack each other forming rigid clusters. Thus, the option which better explains the effect is D. maintain membrane fluidity through its disruption of fatty acid packing.

How did G1 and G2 get there names
Alborosie
Figure 1.1 shows its four main stages:gap 1, synthesis, gap 2, and mitosis.Gap 1, synthesis, and gap 2 together make up what is called interphase. The stages of the cell cycle get their names from early studies of cell division. ... They also carry out cell-specific functions in G1 and G2.

How does a prokaryotic cell such as bacteria differ from eukaryotic cells?
LenKa [72]

Answer:

The main difference is that EUKARYOTIC cells have a nucleus while a PROKARYOTIC cell does not. Instead, prokaryotes have free floating genetic material.

Explanation:

To help you remember this distinction:

EU = YES eukaryotes HAVE a nucleus.

PRO = NO prokaryotes DO NOT have a nucleus.

I hope this helps and good luck with your assignment.
