Exoenxymes function outside cells and endoenzymes function inside cells.  

<h3><u>Explanation:</u></h3>

Enzymes are present everywhere in the body. Some of them are present inside the cells where they are produced and they function. Rest of them are produced inside the cell but are secreted outside so that they can work at their targeted place.  Enzymes of Glycolysis, Krebs cycle, ETS etc , and other house keeping enzymes are functional inside the cells.  

Enzymes required in the digestion like amylase, proteases, maltase, etc are required inside the gastrointestinal tract. So they are secreted form intestinal wall or pancreas into the GIT.

It depends upon the protein and also where the deletion of the single amino acid has occurred. Does ur alter or disrupt an important fundamental function or aspect of the protein such as the capability of substrates to bind to the active site, or is near a region that is primarily for developing the additional structure of the protein and is not as important. In most cases, a single amino acid change will not cause the protein to lose its complete function of be denatured.
