Exercise and a healthy die play an important role in decreasing your chances of developing diseases. However, it should be noted that they are not the sole criteria to completely ensure a disease free life. There are other genetic and external factors which could contribute to developing a serious or non serious diseases. Genetic factors generally indicate inherited diseases from the previous generation in direct genetic line (or blood line). External factors could include environmental hazards (such as exposure to pollution at work or in general), bio-chemical hazards (such as infection by a bacteria or virus or other other chemical toxins or sexually transmitted diseases) and even some trauma leading to a disease. Some of these diseases developed due to external factors are quite serious or even life threatening, e.g  AIDS, Asbestosis, exposure to radio-active emissions. Even some genetic diseases could be very serious. For example, it is not uncommon to inherit cancer or heart disease or diabetes genetically.
